Overview

Transmembrane activator and calcium modulator and cyclophilin ligand interactor (TACI) is a lymphocyte-specific member of the tumor necrosis factor receptor superfamily, encoded by the TNFRSF13B gene. Expressed predominantly in B cells, TACI is a cell-surface receptor that binds the ligands APRIL (TNFSF13) and BAFF (also known as TALL1 or BLYS/TNFSF13B) with high affinity, mediating signal transduction essential for B cell maturation and immune homeostasis. Mutations in TNFRSF13B/TACI are strongly associated with common variable immunodeficiency disorders (CVID), autoimmunity, benign lymphoproliferation, and there is evidence for a role in hereditary susceptibility to hematological malignancies, such as acute myeloid leukemia. TACI influences both the antibody response and the regulation of immune tolerance. Therapeutically, TACI is targeted to modulate aberrant B cell function in various immunological disorders. Safety considerations arise chiefly from its central function in immune regulation, as both loss and gain of function can predispose to immune dysregulation, deficiency, or malignancy[1][2][4][6].
