Transmembrane activator and CAML interactor (TACI), encoded by the TNFRSF13B gene, is a type III transmembrane protein belonging to the TNF receptor superfamily. Primarily expressed on B cells, it regulates B cell activation, differentiation, and immunoglobulin class switching by interacting with ligands like APRIL, BAFF and CAML. Defects in TACI are linked to immunodeficiencies and autoimmunity.
TACI activates NFAT, AP-1, and NF-kappa-B through calcineurin-dependent pathways upon binding to APRIL or BAFF.
