The **TARPγ8-associated AMPA receptor** refers to AMPA-type ionotropic glutamate receptors (AMPARs) complexed with the auxiliary subunit transmembrane AMPA receptor regulatory protein gamma-8 (TARPγ8). AMPARs are tetrameric ligand-gated ion channels mediating fast excitatory neurotransmission in the CNS. TARPγ8 is predominantly expressed in the hippocampus and cortex and is critical for proper surface trafficking, synaptic localization, and functional modulation of AMPARs. This complex is a key regulator of synaptic plasticity and is distinctively implicated in region-specific modulation of excitatory signaling. It is the target of novel antiepileptic drugs that exploit selective negative allosteric modulation at the TARPγ8 interface, offering improved region targeting and minimized side effects compared to non-selective AMPAR antagonists[1][2][3][4][5][8]. Genetic variation in CACNG8 (TARPγ8 gene) is associated with altered expression of this subunit and differences in neuropsychiatric phenotypes and central nervous system disease susceptibility[2][6].
