Transmembrane and coiled-coil domain-containing protein 6 (TMCO6) is a human multi-pass membrane protein encoded by the *TMCO6* gene on chromosome 5 (5q31.3)[1][2]. TMCO6 exists in several isoforms, ranging from 253–499 amino acids, and features two transmembrane domains, Armadillo (ARM) repeat domains, a coiled-coil region, and regions implicated in protein binding and nuclear localization[1][2]. It is predicted to function predominantly in nuclear protein import and as a nuclear import signal receptor, consistent with its nuclear localization sequence, SRP1 domain (importin/karyopherin alpha), and its strong protein binding regions[1][2]. TMCO6 is expressed in several tissues, including the liver and brain, as well as during fetal development, and is evolutionarily conserved in vertebrates but absent in invertebrates and lower organisms[1][2][8]. Experimental and predicted functions also indicate roles in cytoskeletal regulation and possibly ciliogenesis, especially in ependymal cells[7]. No currently known drugs or safety concerns are documented for TMCO6, and there is no current evidence that it is a direct therapeutic target or that it interacts with approved pharmaceuticals[2][4]. Disease associations are limited and largely predicted; TMCO6 has been mentioned in context with mitochondrial complex I deficiency, nuclear type 13, but its mechanistic involvement in disease is unclear[2].
