Overview

TMIGD1 is a highly conserved cell adhesion protein with two immunoglobulin-like domains in its extracellular region, a single transmembrane domain, and a short intracellular tail[1][2][4]. It is predominantly expressed in renal tubular epithelial cells, where it regulates cell–cell adhesion, cell morphology, migration, permeability, and survival under stress[1][4]. TMIGD1 acts as a tumor suppressor in renal cell carcinoma and possibly other epithelial cancers, being downregulated in tumors and upregulated by specific transcription factors such as C/EBPβ[2]. TMIGD1 also protects epithelial cells from oxidative damage and nutrient deprivation, with its loss associated with increased cell death and permeability[1][2][4]. No drugs directly targeting TMIGD1 are reported, but its function and expression status suggest its possible future therapeutic and biomarker relevance in cancer and kidney diseases.
