Transmembrane and tetratricopeptide repeat-containing protein 3 (TMTC3) is an integral endoplasmic reticulum membrane protein with multiple transmembrane domains and large clusters of tetratricopeptide repeats (TPR) in its luminal C-terminal region. TMTC3 acts as a protein O-mannosyltransferase, catalyzing the addition of O-linked mannose glycan to cadherin family proteins, particularly E-cadherin, which is essential for cell–cell adhesion. TMTC3 supports cellular adherence and embryonic gastrulation, and its loss of function or mutations are strongly associated with Cobblestone lissencephaly and other neuronal migration disorders[1][2].
Drugs targeting TMTC3 would theoretically act by modulating O-mannosyltransferase activity, thus impacting glycosylation of cadherins and cell adhesion[1][2]
