Transmembrane BAX inhibitor motif-containing protein 1 (TMBIM1) is a 34.6 kDa integral membrane protein featuring seven transmembrane domains and predominantly localized to endosomal and lysosomal membranes[1]. It plays an essential role in regulating apoptosis through death receptor binding, notably suppressing Fas-mediated apoptosis and maintaining calcium homeostasis[1][4][6]. TMBIM1 is highly expressed in cardiac, skeletal muscle, and brain tissues, but absent in thymus, spleen, or testis[1][7]. It has been identified as a negative regulator of the extrinsic apoptotic pathway and a participant in intracellular stress signaling via inhibition of the p38/MAPK cascade, especially in glioblastoma where its upregulation correlates with increased proliferation, reduced apoptosis, and resistance to temozolomide[2]. TMBIM1 is implicated in other diseases such as nonalcoholic steatohepatitis, where it contributes to attenuation of liver inflammation and fibrosis by promoting lysosomal degradation of TLR4, and in cardiovascular pathology where it protects against aortic dilation and cardiac hypertrophy via negative regulation of matrix metalloproteinase-9 (MMP9) and TLR4 signals[1]. No direct therapeutics are currently approved for TMBIM1, but it represents a promising drug target and diagnostic biomarker in oncology and metabolic/cardiovascular diseases[1][2][9].
