Transmembrane channel-like protein 3 (TMC3) is a member of the TMC protein family, structurally characterized by multiple transmembrane domains and conservation across diverse animal species[1][2][3]. TMC3 is predicted to function as a mechanosensitive monoatomic ion channel, involved in monoatomic ion transmembrane transport, and may modulate membrane excitability[3]. Although TMC1 and TMC2 have established roles in mechanotransduction (notably in hearing), TMC3's physiological functions in mammals are less defined but have been experimentally shown to confer mechanosensory responses when ectopically expressed in model organisms such as Caenorhabditis elegans, suggesting potential involvement in sensory transduction[2]. Some evidence indicates TMC3 may be expressed in subsets of sensory neurons in mice and could play a role in mechanosensation or thermosensation, but direct evidence of its precise function and relevance as a therapeutic target remains lacking[2]. No clinically validated drugs or biomarkers are currently associated with TMC3; its designation as a direct therapeutic target is not supported by current knowledge[1][3].
