Overview

Transmembrane channel-like protein 5 (TMC5) is a member of the TMC (transmembrane channel-like) family, predicted to function as a mechanosensitive monoatomic ion channel. It is classified as a multi-pass membrane protein expressed in various tissues. TMC5 is notably overexpressed in multiple human cancers, including prostate and gastric cancer, and is implicated in promoting tumor cell proliferation, migration, invasion, and cancer progression, primarily through regulation of the cell cycle and activation of oncogenic pathways such as PI3K/AKT, RAS/MAPK, and TSC/mTOR. Its overexpression correlates with poor prognosis in certain cancers, and TMC5 knockdown suppresses cancer cell growth and increases sensitivity to some chemotherapeutic agents (e.g., 5-fluorouracil). TMC5 is thus considered a potential therapeutic target and a biomarker for diagnosis and prognosis in oncology research[1][2][3][4].
