Overview

Transmembrane emp24 domain-containing protein 10 (TMED10) is a type I transmembrane member of the EMP24/GP25L/p24 family, characterized by a GOLD domain and mainly localized to the plasma membrane and Golgi cisternae[1][2][3]. TMED10 primarily functions as a cargo receptor and regulator in vesicular trafficking, mediating protein transport between the endoplasmic reticulum (ER), the ER-Golgi intermediate compartment, and the Golgi apparatus via association with COPI and COPII vesicles[3][5]. It facilitates the selection and quality control of secretory and membrane proteins, is involved in the assembly and maturation of vesicle coats, and has a regulatory role in the activity of the gamma-secretase complex without affecting its epsilon-secretase activity[1][3]. TMED10 is implicated in the regulation of amyloid precursor protein processing and has been associated with early-onset familial Alzheimer’s disease[1][3][5]. As a member of the TMED/p24 protein family, TMED10 is required for correct embryonic development, likely due to its fundamental role in intracellular protein trafficking and structural organization of early secretory pathway organelles[5]. There are currently no direct drugs or therapies targeting TMED10, and it is not widely used as a biomarker or therapeutic target.
