Overview

Transmembrane p24 trafficking protein family member 8 (TMED8, FAM15B, L10) is a protein-coding gene in humans that has been listed as part of the TMED/p24 protein family. However, according to recent reviews, TMED8 is a GOLD domain-containing protein that, unlike true TMED/p24 family members, lacks a transmembrane domain and certain key conserved cysteine residues. As a result, TMED8 is not considered a bona fide p24/TMED family member and is incorrectly designated as such in several protein databases and literature sources[1][3]. Unlike canonical TMED family members, which are involved in protein sorting and vesicle trafficking in the early secretory pathway, the precise function, disease relevance, and therapeutic potential of TMED8 remain poorly defined. There is no evidence supporting its direct role as a receptor, transporter, or druggable target, and there are no established drugs, mechanisms, biomarkers, or safety issues related to TMED8.

Biological functions

Possibly involved in intracellular protein transport (by analogy to other TMED family members, but evidence in humans for TMED8 is lacking)Quality control in protein sorting (if indeed functional as a TMED, but disputed for TMED8)
03

Disease associations

Other (no clear or specific disease associations established for TMED8 in the literature; broader TMED family members may play roles in cancer, immune response, neurodegenerative diseases, and diabetes, but TMED8’s role is not discussed in these contexts)
