Overview

Transmembrane p24 trafficking protein 1 (TMED1) is a member of the p24 (TMED) family, a group of transmembrane proteins crucial for vesicular trafficking of membrane-bound proteins between the endoplasmic reticulum and the Golgi apparatus. TMED1 acts as a cargo receptor, facilitating the incorporation of secretory proteins into COPI and COPII-coated vesicles and may help with vesicle coat formation. It contributes to secretory pathway integrity and is widely expressed in human tissues. TMED1 has been shown to positively regulate interleukin-33 signaling through interactions with the IL1RL1 receptor and participates in innate immune signaling via the cGAS-STING pathway. Its upregulation is associated with several cancers, especially colorectal cancer, serving as both an oncogene and a poor prognosis biomarker, and knockdown experiments indicate roles in cell proliferation, apoptosis, and cell cycle regulation.
