Transmembrane p24 trafficking protein 3 (TMED3) is a member of the p24 (TMED) protein family, which comprises type I transmembrane proteins essential for vesicular trafficking between the endoplasmic reticulum (ER) and the Golgi apparatus[1][2][4][5]. It is involved in the formation of COPI and COPII-coated vesicles, playing key roles in the selection and transport of cargo proteins, maintenance of the Golgi apparatus, and regulation of early secretory pathway dynamics[4][5]. TMED3 also functions as a quality control receptor by recognizing appropriately folded proteins, and influences various signaling cascades, including the WNT-TCF and IL-11/STAT3 pathways, in a cell context-dependent manner[1][4]. Its role in cancer is complex and may be tumor-type specific: in colorectal cancer, TMED3 suppresses metastasis through WNT-TCF pathway regulation, whereas in hepatocellular carcinoma, it promotes invasion and correlates with poor prognosis via IL-11/STAT3 signaling[1][3][4]. Elevated TMED3 has been proposed as a prognostic biomarker in colorectal cancer and is implicated in the progression of multiple human cancers[1][2][3].
