Transmembrane phosphatase with tensin homology (TPTE) is a member of the PTEN phosphoinositide phosphatase family and is classified as a cancer-testis antigen (CTA) (UniProt: P56180). In healthy individuals, TPTE expression is largely restricted to the testis, specifically within the membranes of spermatocytes (PubMed: 11013253). However, it is frequently overexpressed in various cancers, including melanoma, prostate, and lung cancer, making it a valuable target for immunotherapy (PubMed: 15632181). Structurally, it contains a phosphatase domain and multiple transmembrane regions, though its enzymatic activity is distinct from PTEN and may involve specific phosphoinositide substrates (PubMed: 10531356). TPTE is currently a key target in clinical-stage cancer vaccines, such as BNT111, which uses mRNA to encode TPTE alongside other tumor-associated antigens to stimulate a robust anti-tumor T-cell response (BioNTech, 2024). Its restricted expression profile in normal somatic tissues minimizes the risk of autoimmune toxicity, although potential off-target effects must be monitored. The protein's role in cancer cell signaling and its high immunogenicity make it an ideal candidate for multi-epitope therapeutic strategies. Clinical trials are ongoing to evaluate the efficacy of targeting TPTE in combination with checkpoint inhibitors to enhance the immune system's ability to recognize and destroy malignant cells.
