Transmembrane phosphatase with tensin homology pseudogene 1 (TPTEP1) is classified as a long non-coding RNA transcript derived from the TPTE pseudogene family, lacking protein-coding capacity but exerting functional roles through its RNA sequence. TPTEP1 is notably downregulated in multiple cancers, including lung, glioma, ovarian, and gastric cancer, and acts as a tumor suppressor by inhibiting cell proliferation, cancer stemness, migration, invasion, and promoting apoptosis. Mechanistically, TPTEP1 functions as a competitive endogenous RNA (ceRNA), sponging oncogenic microRNAs (such as miR-328-5p, miR-106a-5p, miR-548d-3p), thereby upregulating tumor suppressor genes (e.g., SRCIN1, MAPK14, KLF9/PER1) and inactivating cancer-promoting pathways (PI3K/AKT, SRC, STAT3, MAPK). While not a traditional drug target (receptor, enzyme, transporter), TPTEP1's expression serves as a biomarker for prognosis and disease monitoring in several cancers.
