Overview

Transmembrane protease serine 11A (TMPRSS11A) is a membrane-bound serine protease belonging to the type II transmembrane serine protease (TTSP) family[1][4][5]. It is involved in proteolysis at the cell surface and is predicted to contribute to physiological processes such as cellular senescence, regulation of cell proliferation, and possibly cell cycle arrest by inducing G1 arrest when overexpressed[1][5]. TMPRSS11A is also positioned at the epithelial cell surface, especially in the airway, where related TTSPs can activate viral proteins and facilitate viral entry, implicating a possible role in the pathogenesis of respiratory virus infections[2]. Disease associations include cancer—particularly its identification as the esophageal cancer-susceptibility gene 1 (ECRG1)—as well as iron-refractory iron deficiency anemia and hypotrichosis[1]. There are currently no approved drugs specifically targeting TMPRSS11A, but general protease inhibitors targeting related TTSPs are considered mechanistically relevant.
