Transmembrane protease serine 2 (TMPRSS2) is a type II transmembrane serine protease that plays a crucial role in both physiological and pathological processes. It facilitates viral entry into host cells by cleaving and activating viral envelope glycoproteins, particularly the spike protein of coronaviruses. TMPRSS2 is also involved in prostate cancer, where TMPRSS2–ERG gene fusions can drive androgen-independent tumor growth. Its expression is regulated by androgenic hormones, making it a potential therapeutic target for both viral infections and prostate cancer.
TMPRSS2 inhibitors block the priming of viral spike proteins, preventing viral entry into host cells.
