Transmembrane protein 101 (TMEM101) is a multi-pass integral membrane protein primarily localized to the plasma membrane and nucleoplasm, with eight predicted transmembrane domains. It is involved in the positive regulation of the canonical NF-kappa-B signaling pathway, and is thought to contribute to cell cycle control, DNA repair, and cellular proliferation. High TMEM101 expression has been observed in several tumor types, most notably hepatocellular carcinoma, where it is associated with poor prognosis and more advanced tumor stage and grade. TMEM101 has been proposed as a diagnostic and prognostic biomarker in liver cancer but is not currently a direct target for any approved therapeutic drugs. Its molecular function and broader disease relevance are under ongoing investigation, particularly related to immune infiltration and tumor progression. TMEM101 may act through modulation of the NF-κB pathway, and its higher levels are linked with increased tumor cell proliferation and decreased anti-tumor immune cell presence in some cancers. No approved pharmacological agents or targeted therapies for TMEM101 are currently documented in clinical or preclinical literature. As a biomarker, TMEM101 has shown diagnostic performance comparable to or superior to some established markers (such as AFP) in hepatocellular carcinoma, but it is not yet widely used in clinical practice. TMEM101 is classified as an "Other" under molecular families due to the lack of evidence for it being a classic receptor, ion channel, enzyme, or transporter.
