Overview

Transmembrane protein 106C (TMEM106C) is an integral protein of the endoplasmic reticulum membrane encoded by the TMEM106C gene in humans. The protein contains a domain of unknown function (DUF1356), spans the membrane twice, and is found to be highly conserved across species. It has no well-characterized biological function but is notable for being overexpressed in a variety of cancer types and has been implicated in conditions such as frontotemporal dementia/ALS and distal arthrogryposis. TMEM106C is not considered a conventional therapeutic target (e.g., receptor, enzyme, transporter), and there are currently no known drugs that specifically interact with this molecule, nor established mechanisms of action or direct safety concerns associated with it. Its overexpression in several tumor tissues suggests potential as a biomarker for certain cancers.
