Overview

Transmembrane protein 114 (TMEM114) is a glycosylated transmembrane protein, highly conserved in vertebrates, and primarily expressed in the lens epithelium of the eye. It is associated with ocular development, with disruption or mutation linked to developmental eye disorders such as congenital cataract and microphthalmia. TMEM114 is structurally distinct from claudin family members and has homology to voltage dependent calcium channel gamma subunits but lacks some of their characteristic domains. Its expression at the plasma membrane is dependent on N-linked glycosylation. The precise cellular function is still under investigation, but it is considered essential for normal lens and eye formation. Key clarification: TMEM114 and Claudin-26 are not the same protein. Past database confusion is corrected by recent biochemical analyses; it is classified as a glycosylated transmembrane protein with significant roles in lens development, not a tight junction claudin nor an ion channel subunit.
