Transmembrane protein 115 (TMEM115) is an integral membrane protein primarily localized to the Golgi complex, where it regulates Golgi-to-endoplasmic reticulum (ER) retrograde transport[1][2][3][4]. TMEM115 contains four candidate transmembrane domains primarily in its N-terminal region and features cytoplasmic N- and C-terminal domains, with the C-terminal region exposed to the cytoplasm[1][2]. Functionally, TMEM115 interacts with components of the conserved oligomeric Golgi (COG) complex and COPI, and it may form homo-oligomers or dimers. The region immediately distal to the fourth transmembrane domain (residues 206–229) is both necessary and sufficient for Golgi targeting[1]. TMEM115 knockdown or overexpression disrupts Golgi-to-ER retrograde transport, altering O-linked glycosylation patterns and potentially impacting glycosylation enzyme distribution via trafficking defects[1][2][3]. TMEM115 expression is dysregulated in some cancers and metabolic disorders, although it is not established as a therapeutic target or known drug target.
