Overview

Transmembrane protein 116 (TMEM116) is an integral transmembrane protein predominantly localized to the endoplasmic reticulum, with some isoforms potentially localizing to mitochondria[2]. TMEM116 is widely expressed in tissues such as lung, kidney, and heart, and is especially upregulated in non-small-cell lung cancer and some other malignancies[1][2][3][5]. In cancer cells, TMEM116 is involved in processes such as cell proliferation, motility, migration, and invasion. Mechanistically, TMEM116 regulates oncogenic pathways including the PDK1-AKT-FOXO3A signaling axis. Knockdown or inactivation of TMEM116 impairs cancer cell growth and metastasis in experimental models, suggesting it acts as a critical integrator of oncogenic signaling in cancer progression and may be a potential therapeutic target in oncology[1]. Broader biological roles may include participation in protein modification and cellular communication, but no direct associations with approved or experimental drugs have been reported to date[5][2][6].
