Overview

TMEM119 is a type-I transmembrane protein prominently and selectively expressed in microglia, the innate immune cells of the CNS, under physiological conditions. TMEM119 is widely used as a marker for microglia in human and murine tissue, facilitating distinction between CNS-resident microglia and blood-derived macrophages. Altered TMEM119 expression – typically decreased in reactive microglia during neurodegeneration and inflammation – may reflect microglial activation states. Its utility as a marker is challenged in disease where expression fluctuates or is present in peripheral tissues. TMEM119 also has non-CNS roles, promoting bone formation, mineralization, and spermatogenesis through interaction with osteogenic pathways (BMP2, ATF4). Currently, it is not known as a direct therapeutic target, but is valuable in research for monitoring microglia in neuroinflammatory and neurodegenerative diseases.
