Transmembrane protein 131-like (TMEM131L) is a largely uncharacterized integral membrane protein encoded on human chromosome 4q31.3, conserved across eukaryotes. It is involved in the negative regulation of canonical Wnt signaling, primarily acting at the level of LRP6 coreceptor degradation, and regulates proliferation of immature thymocytes—a function essential for proper T cell development and immune homeostasis. TMEM131L is expressed highly in immune-related tissues such as lymphocytes and bone marrow, with upregulation observed during acute immune responses and in various cancers, most notably gliomas. Additionally, TMEM131L has been implicated in collagen processing and secretion, indicating a possible role in extracellular matrix regulation. Recent studies have identified TMEM131L as a prognostic biomarker for poor outcome in glioma, suggesting influence over the tumor immune microenvironment and cell proliferation. Despite these associations, there are currently no approved or experimental drugs known to directly target TMEM131L, and its full spectrum of clinical impact remains under investigation. Note: No evidence suggests TMEM131L is a typical pharmacologic "target" (e.g., receptor, enzyme, transporter); it is primarily a membrane protein with regulatory roles and is being explored as a biomarker and for mechanistic insight in cancer and immunology.
