Overview

Transmembrane protein 140 (TMEM140) is a multi-pass membrane protein encoded by a human gene located on chromosome 7q33. Its expression is notably increased in glioma and certain other cancers, where it promotes cell growth, survival, adhesion, migration, and invasion. TMEM140 knockdown induces cell cycle arrest and apoptosis, primarily through dysregulation of adhesion molecules and apoptotic factors. TMEM140 has also been identified as a host factor that impedes herpes simplex virus-1 replication by interfering with crucial viral protein interactions. While no clinical therapeutics or chemical probes specifically target TMEM140 currently, its role in tumor progression and viral infection points to its potential as both a prognostic biomarker and a therapeutic target in oncology and infectious disease research

Mechanism of action

For siRNA/knockdown strategies: Antagonism by silencing the gene, which leads to reduced cell proliferation, adhesion, invasion, and increased apoptosis in glioma cells. For viral infection: TMEM140 competitively inhibits the binding of HSV-1 UL31 to UL34, blocking nucleocapsid egress.
