Overview

Transmembrane protein 143 (TMEM143) is a dual-pass transmembrane protein primarily localized to the mitochondrial membrane, with possible presence in the plasma membrane and other organelles[2][1]. It is highly expressed in human heart and skeletal muscle, and predicted functions include acting upstream in hematopoietic progenitor cell differentiation and serving a linker or scaffold role at organelle contact sites[1][3][4]. TMEM143 is not currently classified as a typical therapeutic target such as a receptor, enzyme, or transporter, nor are there any approved pharmaceutical agents directed at it. Expression and interaction studies suggest a potential, but unproven, role in cancer regulation and lipid metabolism[2]. It has several aliases and is conserved across vertebrates. Due to a lack of clear mechanistic or disease-association evidence, TMEM143 is not considered a therapeutic target at this time[2][4][1].
