Overview

Transmembrane protein 151B is a human protein encoded by the TMEM151B gene, located on chromosome 6 (6p21.1)[1][5][7]. The gene encodes a predicted 566-amino-acid protein with two transmembrane domains and a molecular weight of approximately 61 kDa[1][2]. TMEM151B shows high expression in the brain, especially in the cerebellum, medulla, and olfactory bulb, as well as in the testes[1][3]. The biological function remains unclear, as it is primarily annotated as a membrane-associated protein of unknown function and not linked to any well-defined molecular family such as receptor, transporter, enzyme, or ion channel[2][3][5][7]. There is limited functional data; protein–protein interaction studies indicate a single reported interactor, SREBF2, a transcription factor precursor related to cholesterol biosynthesis[1]. Disease associations are indirect and include links to altered expression in cancers (colorectal and breast), muscle phenotype (lean muscle via GWAS), and ischemic neuropathy[1][7]. There are no known small-molecule drugs or targeted therapies, and no recognized use as a biomarker or noted safety concerns. TMEM151B is classified as a poorly characterized transmembrane protein rather than as a classic therapeutic target (i.e., receptor, transporter, enzyme)[1][2][7].
