Overview

Transmembrane protein 154 (TMEM154) is a single-pass transmembrane protein, predicted to localize in the plasma membrane and cytoplasm, and is encoded by the TMEM154 gene (NCBI Gene ID: 201799)[1][3][4]. Its precise molecular function is currently unknown in any species[2][3]. In humans, TMEM154 mRNA is most abundant in immune-related cells such as CD19+ B cells and CD14+ monocytes[2][4]. Genetic studies highlight TMEM154 as a susceptibility locus for type 2 diabetes in humans, with disease-associated variants linked to altered glucose metabolism or insulin signaling, although specific mechanisms are not defined[1]. In sheep, different TMEM154 protein isoforms confer differential susceptibility to ovine lentivirus (OPPV): the ancestral allele is associated with infection, while a particular mutant allele reduces susceptibility. Complete loss-of-function mutations do not appear to impair survival or reproduction in sheep[2]. No drugs are documented as interacting with human TMEM154, and no mechanism of action or safety concerns are reported in therapeutic contexts. TMEM154 is not currently recognized as a direct therapeutic target (e.g., receptor, enzyme, transporter)[1][2][3][4].
