Overview

Transmembrane protein 158 (TMEM158) is a multi-pass membrane protein, first described as upregulated during RAS-induced senescence in fibroblasts, and also known as Ras-induced senescence protein 1 (RIS1)[1][2][3]. Although initially linked to a tumor suppressor response, TMEM158 is now recognized as playing an an oncogenic role in several solid tumors, including gliomas, ovarian cancer, pancreatic cancer, colorectal cancer, and triple-negative breast cancer. It promotes cancer cell proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT), primarily through activation of the STAT3 and TGF-β signaling pathways[1][2][3]. TMEM158 has been reported to serve as a prognostic biomarker, with high expression levels correlating with poor clinical outcomes across multiple tumor types, and it may play a role in mediating resistance to several cancer drugs. TMEM158 does not currently belong to any major receptor or transport protein superfamily, is not a classic GPCR/ion channel/enzyme, and there are no approved drugs targeting it directly as of the latest reports[1][2][3].
