Overview

Transmembrane protein 170A is a small, conserved human transmembrane protein that localizes to the membranes of the endoplasmic reticulum (ER) and the nuclear envelope. It functions as a key regulator of ER and nuclear envelope morphogenesis by promoting the formation of ER sheets at the expense of tubular ER. TMEM170A concentration influences the morphology of both the ER and nuclear envelope—downregulation induces tubular ER and nuclear envelope expansion, while overexpression promotes ER sheet formation and can inhibit nuclear envelope expansion. TMEM170A interacts antagonistically with RTN4 (a reticulon family protein), balancing the shape of the ER and the organization of the nuclear envelope and nuclear pore complexes. While it is not currently known as a therapeutic target or implicated in specific diseases, its central role in maintaining ER and nuclear envelope architecture is critical for cellular homeostasis[1][2][3][4].
