Overview

Transmembrane protein 174 (TMEM174) is a protein-coding gene highly expressed in the kidney and localized to the apical plasma membrane of renal proximal tubular cells[1][4]. TMEM174 functions as a regulator of phosphate ion homeostasis by modulating the sodium-phosphate cotransporter SLC34A1 (NaPi2a), with its activity influenced by parathyroid hormone (PTH) and fibroblast growth factor 23 (FGF23)[1][3]. TMEM174 also enhances cell proliferation by activating the AP-1 transcription factor, partly through ERK signaling[2][3]. Elevated expression of TMEM174 is associated with certain renal cancers and may play a significant role in the development and progression of kidney cancer[2][3]. Currently, no drugs targeting TMEM174 are reported, and clinical utility as a biomarker or therapeutic target remains under investigation.

Mechanism of action

Not established for drugs due to lack of drug interactions; however, TMEM174 modulates phosphate reabsorption via interaction with NaPi2a and responds to phosphaturic hormones (PTH, FGF23)[1][3].
