Overview

Transmembrane protein 176A (TMEM176A) is a member of the MS4A family of proteins, functioning primarily as an intracellular cation channel localized around the trans-Golgi network[1]. It is broadly expressed in human tissues, with high expression in immune cells, particularly RORγt(+) Th17 cells, suggesting a role in the modulation of intracellular ion flow and influencing immune cell function[1][5]. TMEM176A, together with its homolog TMEM176B, acts to inhibit dendritic cell maturation and antigen presentation, particularly under pathological conditions such as chronic spinal cord injury[1][2][5]. Aberrant expression and methylation of TMEM176A are linked to several cancer types, where it may function as a tumor suppressor by promoting apoptosis or inhibiting migration and invasion; conversely, upregulated TMEM176A is associated with certain lymphomas and lung carcinomas as a potential biomarker[1][2]. Beyond cancer, TMEM176A may also play a role in neuropsychiatric diseases, including schizophrenia[1]. There are currently no known approved drugs directly targeting TMEM176A, and its dual roles in immunity and cancer suggest that therapeutic modulation could have complex effects[1][2][5].
