Overview

Transmembrane protein 178A (TMEM178A) is a membrane-associated protein encoded by the TMEM178A gene on chromosome 2. It acts as a negative regulator of osteoclast differentiation both in basal and inflammatory conditions, by modulating Ca(2+) fluxes induced by TNFSF11 (RANKL), and thus impacts activation of NFATC1, a key transcription factor in osteoclastogenesis. TMEM178A is localized predominantly to the endoplasmic reticulum membrane. There is some predicted involvement in the regulation of cytosolic calcium ion concentration, suggesting a broader modulatory role in intracellular signaling. While associated with bone metabolism and potentially with hereditary angioedema, it is not currently recognized as a therapeutic target or biomarker in clinical oncology, immunology, or metabolic disease
