Overview

Transmembrane protein 196 is a four-pass transmembrane protein primarily localized in the cytoplasm. It acts as a tumor suppressor in lung cancer by inhibiting cell proliferation, migration, invasion, and metastasis. TMEM196 exerts its effects by repressing the Wnt/β-catenin pathway, leading to cell cycle arrest and increased apoptosis. Its reduced expression, often via promoter hypermethylation, is associated with poor clinical outcomes and aggressive tumor behavior. Genetic polymorphisms in TMEM196 are also implicated in respiratory diseases such as asthma, particularly in the context of NSAID-exacerbated respiratory disorders. TMEM196 is considered a promising diagnostic and prognostic biomarker in cancer, but currently lacks known direct drug interactions
