Overview

Transmembrane protein 198 (TMEM198) is a seven-transmembrane protein located at the plasma membrane and in cytoplasmic vesicles. It acts as a membrane scaffold, directly associating with the coreceptor LRP6 to recruit casein kinase family proteins, which is critical for phosphorylation of LRP6 and effective activation of Wnt/β-catenin signaling. TMEM198 is indispensable for both basal and casein kinase 1-induced LRP6 phosphorylation, facilitating signal transduction upon Wnt stimulation. It is expressed broadly during embryogenesis and is essential for processes such as neural crest formation and antero-posterior patterning. While TMEM198 shows molecular associations with diseases like rectosigmoid cancer, there is currently no evidence supporting its use as a direct therapeutic target or as a biomarker in clinical practice[1][2][3].
