Transmembrane protein 201 (TMEM201), also known as NET5, SAMP1, or Ima1, is a conserved inner nuclear membrane protein characterized by five transmembrane segments. In human cells, TMEM201 localizes predominantly to the inner nuclear membrane, where it interacts with the linker of nucleoskeleton and cytoskeleton (LINC) complex via its N-terminus and other nuclear envelope components such as Lamin A/C and SUN2. TMEM201 modulates nuclear-cytoskeletal interactions, cell motility, centrosome positioning, and chromatin organization. It is essential for endothelial cell migration and angiogenesis in both in vitro and in vivo models, and its deficiency results in impaired vessel formation and reduced cell movement. Aberrant expression of TMEM201 has been implicated in tumor progression, highlighting its potential as a marker related to cancer cell migration. TMEM201 is not a classical drug target, but its functional role in cell migration and nuclear architecture makes it of emerging interest in the study of vascular biology and tumor metastasis.
