Transmembrane protein 207 (TMEM207) is a membrane-bound protein localized predominantly in early endosomes with an N-terminal signal peptide and a cytoplasmic C-terminus. It has been implicated in the regulation of cellular biosynthesis, adaptive immune response, and, most notably, in various cancers. TMEM207 can bind to and block the activity of the tumor suppressor WWOX, promoting tumorigenesis, invasion, and aerobic glycolysis in certain carcinoma cells. Its physiological expression is comparatively restricted (mainly in the kidney), but it is overexpressed in multiple tumor types, such as gastric signet-ring cell carcinoma, oral squamous cell carcinoma, and cutaneous adnexal tumors, and has also been postulated to be involved in intestinal immunity and hypoxia response. TMEM207 does not currently have known direct drug interactors or established mechanisms of action for therapeutics but is being evaluated as a prognostic biomarker in certain cancers and as a potential new therapeutic target[1][3].
