Transmembrane protein 209 (TMEM209) is a protein located primarily at the nuclear envelope and potentially the Golgi apparatus, with a structural role as an integral membrane protein[2][3][5]. TMEM209 contains two predicted transmembrane domains and has been implicated as a nucleoporin or nuclear envelope protein interacting with core nuclear pore complex (NPC) components, such as NUP205, POM121, and Nup210[2][3]. It is thought to contribute to nuclear integrity, chromatin organization, and regulation of gene expression[2]. Experimental depletion of TMEM209 results in alterations to nuclear envelope morphology and impairs cell cycle progression, particularly during the S phase[2]. Increased expression and interaction with NUP205 have been associated with lung cancer cell proliferation, suggesting a possible role in tumorigenesis[9][3]. TMEM209 is poorly characterized, with no known drug interactions or established biomarkers, and is not currently recognized as a direct therapeutic target.
