Transmembrane protein 217 (TMEM217) is a multi-pass transmembrane protein encoded by the TMEM217 gene located at 6p21.2 on chromosome 6[2][3]. The canonical protein consists of 229 amino acids and is predicted to contain four transmembrane domains and a domain of unknown function (DUF4534)[2]. TMEM217 is mainly expressed in the lymphatic system, vascular/arterial endothelial tissue, bladder, as well as in other tissues such as connective tissue, liver, mammary gland, testis, and cervix[2]. Functional studies suggest TMEM217 may be involved in cell proliferation and death, with a potential association with cytoskeletal regulation[2][4]. Elevated TMEM217 expression has been linked to several diseases, including acute myeloid leukemia (as an unfavorable prognostic factor and regulator of proliferation and apoptosis in AML cell lines), bladder cancer, lymphoma, Alzheimer’s disease, and diabetic retinopathy[2][4]. No approved drugs are currently known to target TMEM217, and its molecular function remains largely uncharacterized.
