Overview

Transmembrane protein 220 antisense RNA 1 (TMEM220-AS1) is a cytoplasmic long non-coding RNA that is endogenously expressed at low levels in hepatocellular carcinoma (HCC), where its downregulation correlates with poor prognosis and increased tumor invasiveness[1][2][3]. TMEM220-AS1 executes its tumor-suppressive function by transcriptionally enhancing the expression of TMEM220 (a membrane protein), inhibiting the Wnt/β-catenin signaling pathway, and acting as a competing endogenous RNA (ceRNA) that sponges oncogenic microRNAs, notably miR-484, to protect tumor-suppressive genes (e.g., MAGI1)[1][2][3]. Experimental overexpression of TMEM220-AS1 in tumor models limits proliferation, migration, invasion, EMT, and metastasis, with strong implications for its potential as a biomarker or therapeutic candidate in HCC[1][2][3].
