Overview

**Transmembrane protein 225B (TMEM225B)** is a poorly characterized multi-pass transmembrane protein. Closely related animal orthologs, such as rat TMEM225, are expressed specifically in the testis and are associated primarily with germ cell lineages, showing increased expression during adult spermatogenic stages[1][2]. The rat and mouse orthologs are composed of four predicted transmembrane domains and localize mainly to regions near the nuclear envelope and acrosomal membranes rather than the cell surface[1][2]. In mice, TMEM225 has been shown to interact with protein phosphatase 1γ2 (PP1γ2) and to potentially modulate its activity through a conserved RVxF motif, suggesting a possible regulatory role in sperm maturation and function[2]. There is no evidence directly characterizing human TMEM225B's physiological or pathological roles, nor is it recognized as a therapeutic drug target or established biomarker in current biomedical literature[1][2].
