Overview

Transmembrane protein 243 (TMEM243) is a small protein of 118 amino acids predominantly localized to the membrane, with specific accumulation in the mitochondria[1][10]. TMEM243 is overexpressed in certain taxol- and doxorubicin-resistant cancer cell lines, where its induction corresponds to drug exposure and is co-expressed with MDR1, a well-established drug efflux transporter. While TMEM243 does not belong to major drug target families such as G-protein coupled receptors, ion channels, enzymes, or nuclear receptors, its role in mitochondrial function and drug resistance pathways suggests potential relevance in cancer biology, but it is not currently a known therapeutic target[1][2][10]. There are no approved drugs that directly inhibit or modulate TMEM243, nor is it established as a validated biomarker or drug target.
