Overview

Transmembrane protein 25 (TMEM25) is a single-pass transmembrane glycoprotein belonging to the immunoglobulin superfamily. It localizes to tight junctions in epithelial cells, where it interacts with the polarity protein Par3 via its C-terminal cytoplasmic tail and with claudins-1 and -2 via its extracellular domain. TMEM25 negatively regulates the assembly of tight junctions by inhibiting both cis- and trans-oligomerization of claudins, thereby modulating epithelial barrier function[1][2][3]. In neuronal tissues, TMEM25 is involved in modulating degradation of the NMDA receptor subunit GRIN2B and in regulating neuronal excitability, as well as controlling protein stability[4]. TMEM25 is highly expressed in neural progenitor cells and is implicated in cortical development by promoting progenitor expansion; knockdown disrupts extracellular signaling (reducing Akt pathway activity) and inhibits the cell cycle[1]. Epigenetic silencing of TMEM25 by CpG island hypermethylation is associated with colorectal cancer, suggesting a tumor suppressor function, and TMEM25 is also a candidate prognostic biomarker and therapeutic target in clear cell renal cell carcinoma[1].
