Overview

Transmembrane protein 26 (TMEM26) is a multi-pass transmembrane protein encoded by the TMEM26 gene on human chromosome 10, with conservation across mammalian species[1][3]. The protein possesses eight hydrophobic transmembrane domains, multiple hydrophilic cytoplasmic and extracellular domains, and exists in several N-glycosylated isoforms[1][2]. TMEM26 is expressed in many tissues, including the retina—primarily in the inner and outer plexiform layers—breast cancer cells, embryonic mesenchyme, and airway and adipose tissues[1][2][3]. In adult mice and humans, TMEM26 serves as a distinctive cell surface marker for beige (brite) adipocytes and is involved in the browning of white adipose tissue, relevant to metabolism and obesity studies[1][2]. It is also weakly associated with disease phenotypes such as primary open-angle glaucoma, COPD, and various cancers, though it is not a primary driver or established therapeutic target in these conditions[1][3]. In breast cancer, its glycosylation status affects cellular localization and protein stability, but no approved drugs directly target TMEM26[2][4]. Overall, while TMEM26 plays roles in development and cell metabolism, its precise biological function and role as a direct therapeutic target remain unclear and appear to be context-dependent.
