Transmembrane protein 8B (TMEM8B) is a plasma membrane-spanning protein encoded by the TMEM8B gene located on chromosome 9p13.3, expressed most highly in brain, ovaries, prostate, placenta, and pancreas. It is involved primarily in cell-matrix adhesion, inhibition of cell proliferation, and regulation of cell cycle progression by negatively modulating the EGFR pathway. TMEM8B is downregulated in several cancers, particularly nasopharyngeal and colorectal carcinoma, suggesting a tumor suppressor role[1][6]. Isoform TMEM8B-a is subject to rapid proteasomal degradation, mediated by ezrin, which can be inhibited by drugs such as MG-132 and bortezomib[2]. Protein-protein interaction partners include EGF and ATXN1L, linking TMEM8B to both oncogenesis and neurodegeneration. Pathogenic variants may cause chronic pain disorders, most notably erythromelalgia. TMEM8B has multiple mRNA splice variants and a conserved structure with seven membrane-spanning domains and leucine-rich regions[1].
