Overview

Transmembrane serine protease 12 is a type II transmembrane, trypsin-like serine protease and member of the TMPRSS protein family, predominantly expressed in spermatocytes, spermatids, and the acrosome of mature sperm within the testis. TMPRSS12 is essential for proper sperm motility and migration through the uterotubal junction, primarily by regulating the processing of membrane proteins such as ADAM3. Knockout mouse models demonstrate that loss of TMPRSS12 leads to male infertility due to defective meiosis, impaired mitochondrial development in spermatids, reduced sperm motility, and failure of sperm to traverse the UTJ. TMPRSS12 also functions as an activating protease for certain viral fusion proteins (e.g. subtype B avian metapneumovirus), suggesting a potential role in infectious disease mechanisms. The protein comprises a transmembrane domain and a C-terminal serine protease domain facilitating substrate cleavage via a canonical catalytic triad. While no specific drugs target TMPRSS12 in clinical practice, its unique expression and functional role in the testis make it a candidate for research into contraceptive agents or infertility treatments.

Mechanism of action

Drugs (if developed) would likely act by inhibiting the serine protease activity, thereby blocking proteolytic processing of downstream substrates such as ADAM3 or viral fusion proteins. The catalytic triad is the mechanism for proteolysis.
