Overview

Transmembrane serine protease 7 (TMPRSS7, also known as matriptase-3) is a membrane-anchored serine protease belonging to the type II transmembrane serine protease (TTSP) family[1][2][5][6]. It is composed of several protein domains, including a transmembrane domain and a C-terminal serine protease domain. TMPRSS7 is phylogenetically conserved across vertebrates and is predominantly expressed in brain, skin, reproductive, and oropharyngeal tissues[2]. Functionally, TMPRSS7 catalyzes protein and peptide hydrolysis with a preference for arginine in the substrate's P1 position and participates in proteolytic processes at the cell surface. TMPRSS7 genetic variants have been linked to increased risk and poorer prognosis in breast cancer, suggesting a role in modulating tumor microenvironment and cancer progression[1]. TMPRSS7 is therefore considered a potential biomarker and putative therapeutic target in oncology, though targeted drugs have not yet been reported.
