Transport and Golgi organization protein 6 homolog (TANGO6) is a transmembrane, coiled-coil domain-containing protein encoded by the TANGO6 gene in humans[1][13]. TANGO6 localizes primarily to the cytoplasm and perinuclear area, associating dynamically with COPI vesicles and the Golgi apparatus throughout the cell cycle[2]. It mediates retrograde transport from the Golgi to the endoplasmic reticulum (ER) and is implicated in the nuclear transfer of essential factors, such as RNA polymerase II subunit B (RPB2), thereby regulating cell proliferation and cell cycle progression, notably the G1 phase[2]. Genetic loss or overexpression affects hematopoiesis and cell expansion but does not currently classify TANGO6 as a therapeutic target or receptor. Its exact molecular functions are still being clarified, and there is no evidence to date for direct clinical utility as a drug target or biomarker[2][3][4][11].
