Overview

Tribbles homolog 1 (TRIB1) is a highly conserved pseudokinase that acts as an adapter/scaffolding protein rather than a phosphotransferase, due to its non-functional ATP-binding domain and atypical SLE motif[1][2][3][4][6]. TRIB1 regulates cellular processes by binding MAPK pathway components and modulating the activity and degradation of critical transcription factors, notably C/EBPα. It is widely expressed and plays major roles in hematopoietic cell differentiation, macrophage polarization, immune response, lipid metabolism, and carcinogenesis[1][2][3][6]. TRIB1's ability to modulate signaling and protein stability makes it a significant therapeutic target and potential biomarker in oncology, metabolic syndrome, cardiovascular disease, and immune regulation, although no specific drugs directly targeting TRIB1 are approved thus far.

Mechanism of action

Potential inhibitors would likely disrupt TRIB1's scaffold/adaptor function and its interaction with substrates (such as C/EBPα) or effectors (such as MAPK, MEK1/2, Akt, JAK/STAT, COP1).
