Trichodiene synthase is a sesquiterpene cyclase enzyme primarily found in filamentous fungi, such as those in the genus Fusarium [1]. It catalyzes the complex cyclization of farnesyl pyrophosphate (FPP) to form trichodiene, which is the first committed step in the biosynthesis of trichothecene mycotoxins [2]. These mycotoxins, including deoxynivalenol (DON) and T-2 toxin, are potent inhibitors of eukaryotic protein synthesis and represent significant contaminants in global food supplies [3]. Exposure to these toxins leads to mycotoxicosis in humans and livestock, characterized by symptoms such as vomiting, immunosuppression, and gastrointestinal distress [4]. Consequently, Trichodiene synthase is a critical target for the development of antifungal agents and agricultural strategies aimed at mitigating toxin production in crops [2]. Research into this target involves the design of transition-state analogs and competitive inhibitors that mimic the carbocation intermediates of the cyclization reaction [1, 2]. While not a target for treating endogenous human diseases, its role in food safety and preventing fungal pathogenesis makes it a high-priority molecule in agricultural biotechnology and public health [3, 4].
